Estamos interessados em identificar a dinâmica das alterações de Ca2+ nos parasitas vivos da malária dentro das células hospedeiras. Descobrimos que, uma vez que o Plasmodium infecta a hemácia, este cria em torno de si um microambiente rico em Ca2+ necessário para explorar a via de sinalização do Ca2+. Um regulador chave da sinalização do Plasmodium IP3 - Ca2+ é o hormônio hospedeiro melatonina. Geramos um sensor de Ca2+ o PfGCaMP3, que permite a obtenção de imagens das mudanças de concentração do íon no interior do parasita.

Buscamos entender o papel dos candidatos a GPCR (G protein coupled receptor), ou receptor serpentino do Plasmodium. Nosso laboratório identificou a primeira proteína do tipo GPCR like no parasita da malária humano P. falciparum. O receptor serpentino PfSR25 detecta uma mudança na concentração de potássio que é normalmente encontrada dentre os ambientes interno e externo das células. Os parasitas nocaute para o receptor PfSR25 são mais suscetíveis aos antimaláricos cloroquina , piperaquina, lumefantrina e para sobrevivência ao estresse.

Nosso objetivo é descobrir como a sinalização por sinais externos afeta e molda as alterações na transcrição de genes e a atividade do sistema ubiquitina-proteassoma (UPS) durante o ciclo celular do parasita.

Outra linha de pesquisa do laboratório inclui a triagem de compostos sintéticos e naturais com foco no desenvolvimento de novos medicamentos para o tratamento da malária. Para isto, utilizamos triagem em larga escala e a técnica de FACS.
